Genstruct Selected to Present Causal Mechanisms for Diabetes at ADA Annual Meeting

Cambridge, Mass. — May 27, 2004 — Genstruct Inc., a knowledge-driven discovery company, will present findings from one of its discovery programs at the 64th annual meeting of the American Diabetes Association, being held in Orlando, Florida from June 4 - 8, 2004. Jack Pollard, Ph.D., lead scientist for the Metabolic Disorders Program, will outline the programs findings that describe both probable molecular causes and intervention points to treat type 2 diabetes.

To define the molecular causes of disease, Genstruct utilized its Molecular EpistemicsTM discovery platform to build and interrogate a Diabetes Knowledge AssemblyTM model using computer-aided causal reasoning. The model includes the knowledge of insulin action, cell signaling, vesicle trafficking, metabolism, and gene regulation. It is the largest and most complete computable model of diabetes knowledge ever assembled.

Dr. Pollard currently directs Genstruct's research efforts in metabolic disorders. Prior to his tenure at Genstruct, Dr. Pollard was director of Informatics Research at 3rd Millennium, where he led research efforts in gene expression analysis and biopathways reconstruction. Dr. Pollard has authored articles in the areas of biochemistry as well as informatics and has made significant contributions to the fields of semantic integration and computer-aided reasoning.

"We utilize our Molecular Epistemics platform to define biological mechanisms for disease. Jack's broad experience in scientific management and development has allowed us to accelerate our metabolic disorders program," said Keith O. Elliston, Ph.D., president and CEO of Genstruct. "The findings that Jack will present demonstrate our ability to distinguish molecular causes from the well characterized consequences of insulin insensitivity in type 2 diabetes."

Genstruct deploys its Molecular Epistemics discovery platform independently and in partnerships with pharmaceutical companies to define biological mechanisms that streamline drug discovery and improve clinical trial design and implementation. The platform enables the development of logical models of biological systems that uncover and define compound mechanisms of action and disease mechanisms. Genstruct has ongoing research programs in prostate cancer and type 2 diabetes mellitus, and is focused primarily on the disease areas of oncology, metabolic disorders, and inflammation. The company believes that the identification of mechanisms and biomarkers in these areas will ultimately lead to improved leads, optimal clinical trial design and execution, and faster FDA product approval.

About Genstruct
Genstruct is a knowledge-driven discovery company focused on pioneering new approaches to the understanding and treatment of complex diseases (i.e. oncology, metabolic disorders, inflammation). Genstruct has developed a platform for the discovery of compound mechanisms of action (MOA) and biomarkers called Molecular EpistemicsTM . The platform comprises a suite of patent-pending technologies that model and analyze compound MOA: the Knowledge AssemblyTM process which enables rapid, ongoing construction of an addressable biological knowledgebase; and a flexible system for mining and refining the knowledgebase, the Epistemics EngineTM system for mechanism discovery and validation. Genstruct engages in discovery and development partnerships that apply the Molecular Epistemics platform to accelerate and increase the output of drug discovery and development programs. The Company was founded in 2001 by a team including Flagship Ventures, and is based in Cambridge, Mass. For more information please visit www.genstruct.com.
